"We seek a peaceful transition of power but all options are open," Trump said at a speech to a community of Venezuelan-Americans in Miami, Florida on Monday.
The military leadership has a "vital role" to play, Trump added. The army has so far sided with Maduro, but is believed to be divided.
"If you choose this path you will find no safe harbour, no easy exit and no way out, you will lose everything," Trump said.
Maduro has presided over an economic collapse, with inflation expected to reach 10 million per cent this year and millions of Venezuelans having fled to neighbouring countries.
The US is one of more than 50 countries that have recognised self-appointed interim president Juan Guaido as Venezuela's legitimate leader.
